U.S. Route 129 (US 129) in Florida is a north–south United States Highway. It runs 88 miles (142 km) from Chiefland north to the Georgia State Line in Levy, Gilchrist, Suwannee, and Hamilton Counties.

Between Hildreth and Branford, it follows SR 20, between Branford and Live Oak, it follows SR 249, between Live Oak and Jasper, it follows SR 51, between Hillcoat and Jasper, it follows SR 25, and between Hillcoat and the Georgia border, it follows SR 100.
